Our ssDNA services.

A partnership programme for modified ssDNA. Synthesis services for linear and circular ssDNA. Existing constructs for research.

Modified ssDNA partnership programme.

Internal chemistry at kilobase length

Partner with Moligo to develop long ssDNA with modified nucleotides distributed throughout the strand. Our modification technology is available to selected partners through a co-development programme with an agreed research scope, not an off-the-shelf modification service.

Together, we assess chemistry feasibility, select the base channel and substitution fraction, and agree characterisation and application testing for the construct. Exact positions and regional modification patterns are not controlled by this process.

A matched performance advantage over unmodified long ssDNA has not yet been demonstrated.

Linear ssDNA

Your sequence.
At gene length.

Custom HDR donor templates

Put your donor design into a single strand, with custom sequences up to 10 kb and no added scar sequences.

For research teams developing HDR donors and other sequence-defined tools. In vitro enzymatic synthesis supports long constructs, including challenging GC-rich regions, repeats and secondary structures, subject to construct review.

To order, send your sequence, target length, quantity and intended application. Confirm the construct specification and delivery requirements with our team.

Circular ssDNA

A continuous strand.
No free ends.

Circular donors for non-viral gene insertion research

Combine a custom donor sequence with a closed circular format. Explore topology as a design choice for your gene-insertion workflow.

Moligo’s collaboration with Cellectis evaluated circular versus linear donors in HSPCs. The study supports the circular format under its tested conditions; it does not establish an advantage for chemically modified circular DNA.

Off-the-shelf sequences.

Start with an existing AAV-like or gene-editing construct. Sequences are available on request.

The release criteria below are published in Moligo’s ssDNA service specification. Agree the applicable tests and requirements for your construct before ordering.

Read each result in context.

Sanger alignment is a sequence-identity check, not a per-molecule error-rate measurement. A gel band does not by itself establish a numerical purity percentage or a residual double-stranded DNA detection limit.

Modified DNA needs additional characterisation.

Modification technology is available to selected partners. Within each co-development programme, we agree how to measure incorporated chemistry and composition, and which format and purity assays are appropriate. Nucleotide input ratios alone do not establish product composition.
